Moving to live and work in Portugal, I'm British and my wife is Spanish. Does being married to an EU citizen give me any rights and affect the visa we apply for?

It does, your wife enters as an EU citizen, you accompany her as the family member of an EU citizen which is a much easier process than you applying as a third country citizen.
